Combining the finding of two substrate-binding sites and the transport cycle, we propose that the lipid head group transports through the groove between TMH2 and TMH4, while lipid tails move accordingly on the hydrophobic surface of the TM2 and TM4. This is essentially the same as the previously described ‘credit card model’ (Andersen et al., 2016; Roland and Graham, 2016). The binding of ATP in the E1-ATP state triggers the N domain to move toward and interact with the A domain, thereby stabilizing both domains. However, the A domain must be only partially stabilized and retain some flexibility, because its density is still weak. With the ensuing phosphoryl transfer, the flippases transition from the E1-ATP state to the E1P-ADP state.
Inadequate PC and choline results in impaired VLDL secretion leading to an accumulation of fat and cholesterol in the liver. This accumulation of lipids triggers a cascade of reactions, including mitochondrial dysfunction, generation of reactive oxygen species , lipid peroxidation, DNA damage, inflammation, apoptosis and finally liver damage and disease. Ensuring adequate PC levels is therefore essential in promoting optimal liver health. To examine the effects of lipid loading on gene expression we performed Agilent microarray analysis. Treatment with eLDL as well as with oxLDL led to a strong downregulation of endogenous cholesterol biosynthesis and external uptake. As indicated by Panther pathway overrepresentation analysis this is likely mediated through sterol regulatory element binding proteins .
Knockdown of LPCAT1 reduced lipoprotein secretion but had a very small effect on cellular lipid composition . Previous findings suggested that lipidation of VLDL requires breakdown of stored TAG and transfer of released fatty acids from LDs to VLDL possibly via ER-luminal LDs . Reduced VLDL secretion in HuH7 cells was also observed upon knock-down of ACSL3, and could be rescued by supplementation with PC . Since both ACSL3 and LPCAT1 localize to LDs, this supports the idea that ACSL3 supplies acyl-CoAs to LPCAT1 to generate PC on the LD surface, where about 50% of cellular LPCAT1 are found in HuH7 cells. The influence on lipid secretion may then arise from a changed lipid mobilization from LDs, where an increase in LD surface seems to be connected to increased lipolysis -.
